Effect of Insulin on Function and PDGF Production of Human Umbilical Vein Endothelial Cell


Abstract
Insuhn resistance and hyperinsulinemia are associated to atherosclerosis and are considered as one of independent risk factors in the pathogenesis of atherosclerosis. The effect of insulin on the endothelial cell in the pathogenesis of
atherosclerosis
was not fully elucidated. In this study, the effect of insulin on the endothelia cell function was observed in vitro human umbilical vein endothelial cell(HUVEC) culture condition.
The growth of cells, H3-thymidine incorporation, cellular regeneration and PDGF production by HUVEC at various insulin concentration were measured.
After incubation of 9 days at insulin concentrations of 10, 100, 300, 500 and 1000uU/§¢. Ratios of cell growth were significantly increased at higher insulin concentrations(p<0.05). However, the grwoth rate at glucose concentration of 300mg/§£was
lower
than that at 100mg/§£ at each insulin concentrrations(p>0.05). H©ø-thymidine incorporation into DNA were also stimulated by increasing concentration of insulin, maximum stimulation occuring at 300uU/§¢of insulin concentration (p<0.05) and glucose
concentration of 300mg/§£had lesser effect on DNA synthesis in endothelial cells(p>0.05).
The ratio of migration distance and labelling index at wound edges and cytotoxicity of endothelial cells were not significantly different at various insulin concentration. The production of PDGF by endothelial cells were significantly increased
according to insulin concentrations(p<0.05).
These results showed that the cell growth, thymidine incorporation and PDGF production on human umbilical vein endothelial cell were increased at higher insulin concentrations but cell function such as regeneration and cytotoxicity were not
changed
by
insulin concentrations. We suggested that higher insulin concentrations may be an important role in the pathogenesis of atherosclerosis on human umbilical vein endothelial cells.
